人參皂苷Rh2在體外對大腸直腸癌細胞的毒性作用

人參皂苷Rh2(Ginsenoside Rh2)是人參的主要活性成分之一。我們使用MTT方法以Rh2分別對6株大腸直腸癌細胞株(LoVo、HCT-15、SW-480、SW-620、 SHT-1及SHH-1)及人類正常成纖維母細胞進行生長抑制試驗。其IC50值分別為18.2±0.3、13.6±1.2、13.4±0.5、 12.1±0.6、13.8±0.2、19.9±0.4及52.2±2.3μg/ml,以成纖維母細胞株的IC50作為選擇指標,6株大腸直腸癌細胞 株:LoVo、HCT-15、SW-480、SW-620、SHT-1及SHH-1的選擇指標分別依序為2.9、3.8、3.9、4.3、3.8及 2.6。經過統計學的分析,Rh2對於東西方人種大腸直腸癌細胞株的IC50並無統計學上的差異,但對於較惡性的細胞具有良好的細胞抑制能力。並將 LoVo細胞經過Rh2處理24小時後,以細胞死亡分析測定,證實Rh2誘發大腸直腸癌細胞發生凋亡。顯示Rh2具有成為大腸直腸癌新一代抗癌藥物的潛 力。

Recently, it has been reported that Rh2 has growth-inhibiting effect on various malignant tumor cells. The present study used MTT test to investigate the cytotoxic effects of Rh2 on six human colorectal cancer cell lines and normal skin fibroblasts were used as the control. Rh2 showed dose-dependent cytotoxic effects on colorectal cancer cells. The IC50 value of Rh2 was 52.2±2.3 μg/ml for normal human fibroblast, 18.2±0.3、13.6±1.2、13.4±0.5、12.1±0.6、13.8±0.2 and 19.9±0.4 μg/ml for LoVo、HCT-15、SW-480、SW-620、SHT-1 and SHH-1 cells, respectively。Rh2 was demonstrated inducing apoptosis in colorectal cancer cells.Our results indicate that Rh2 might be a promising novel agent for the treatment of colorectal cancer.
